I usually pour the Amp powder on the water. It dissolve very well. pouring water on the powder makes it unsoluble. and also I have seen some protocols using half water and half Et-OH.

Sodium ampicillin is fairly soluble in water and should be fine at that concentration. However ampicillin trihydrate or ampicillin anhydrous is much much less soluble, I think the limits are around 5-10 mg /ml. I'm guessing you are not using the sodium salt so yes if you want that concentrated of a stock solution you need to add NaOH.

Ampicillin is (slightly) soluble in water, you should be able to achieve 100 mg/ml, thats what our lab used for stocks, however Sigma here: https://www.sigmaaldrich.com/content/dam/sigma-aldrich/docs/Sigma/Product_Information_Sheet/a9393pis.pdf is recommending a 50 mg/ml solution in water, so try that.
